Background
IC50: mababang saklaw ng nanomolar para sa pagsugpo ng lahat ng mga kasapi ng pamilya TRK
Ang LOXO-101 ay isang tropomyosin receptor kinases (TRK) na inhibitor.
Ang pamilya TRK ng mga neurotrophin receptor at ang kanilang mga neurotrophin ligands ay kinokontrol ang paglago ng neuron, pagkita ng kaibhan at kaligtasan.
Sa vitro: Sa nakaraang pag-aaral, ang LOXO-101 ay sinusuri para sa off-target na kinase na pagsugpo ng enzyme laban sa isang panel ng mga non-TRK kinase sa isang konsentrasyon ng 1,000 nM at ATP na konsentrasyon sa paligid ng Km. Ipinakita sa mga resulta na ang LOXO-101 ay may higit sa 50% na pagsugpo para sa isang di-TRK kinase lamang, na kung saan ay ang TNK2 na may IC50 na 576 nM. Bukod dito, walang kaugnay na pagsugpo sa hERG o matagal na pagmamasid ng QT [1].
In vivo: Natuklasan ng pag-aaral ng hayop na ang LOXO-101 ay nakapagpigil sa paglago ng vivo tumor. Ang Athymic nude mice na iniksiyon ng mga cell ng KM12 ay binibigyan ng pasalita ng LOXO-101 araw-araw sa loob ng 2 linggo, at sinusunod ang dosis na nakasalalay sa pagsugpo ng tumor, na nagpapahiwatig ng kakayahan ng LOXO-101 na pigilan ang paglago ng in-vivo tumor [1].
Pagsubok sa klinikal: Ang isang multicenter phase I na dosis-escalation na pag-aaral sa mga pasyente na may advanced solid tumors (ClinicalTrials.gov no. NCT02122913) ay isinasagawa noong 2014 upang suriin ang kaligtasan at PK ng LOXO-101.
